Descrizione sintetica in italiano | For this specific project the objective is the determination of the structure of three flagellar proteins, two FlgE's and one FlgK, and possibly the complex among them. In parallel, we will try to Obtain the structure of the assembly of the organization of FlgE in the filament by cryo-EM, similarly to the case of Salmonella sp. FlgE. In H. pylori, two genes, both coding for FlgE, are present: HP0908 and HP0870. The crystal structure of HP0908, Whose data are Already available, will be refined and completed. FlgK. FlgK from H. pylori (HP1119) has Already Been Expressed. Despite it presents some problems of stability, attempts to grow crystals will be performed. In Additions, FlgL will be cloned and Expressed in order to test The Possibility of growing crystals of the complex. Finally, EM studies of complexes or aggregates of FlgE and FlgK and FlgL will be performed |
