| Descrizione sintetica in italiano | Il programma di ricerca prevede l'espressione in forma ricombinante e la purificazione di proteine coinvolte nel metabolismo del DNA nel patogeno umano Mycobacterium tuberculosis con l'obiettivo di effettuare studi di interazione proteina-proteina e proteina-ligando utilizzando diversi metodi biofisici tra cui la spettrofotometria, la risonanza plasmonica di superfice e la cristallografia a raggi-X per l'analisi strutturale. Il progetto si prefissa di analizzare la cinetica di interazione di complessi proteici attivi nel metabolismo del DNA e di avviare un percorso sperimentale su base razionale per individuare molecole in grado di interferire con i processi metabolici a carico del DNA con lo scopo di sviluppare agenti ad attività anti-micobatterica. |
| Descrizione sintetica in inglese | The research program focuses on the recombinant expression and purification of proteins involved in DNA metabolism in the human pathogen Mycobacterium tuberculosis with the aim of carrying out protein-protein and protein-ligand interaction studies using various biophysical methods including spectrophotometry, surface plasmon resonance and X-ray crystallography for structural analysis. The project aims to analyse the interaction kinetics of protein complexes active in DNA metabolism and to initiate a rationally based experimental pathway to identify molecules capable of interfering with DNA metabolic processes with the aim of developing agents with anti-mycobacterial activity. |
