Descrizione sintetica in italiano | Normal/immune cell interactions are critical for several pathogenetic mechanisms. The full lists of genomic loci, mutations, and epigenetic alterations affecting immunotherapies are not yet known. Immunotherapy is highly effective in immunologically 'hot' tumors; however, new personalized chemo-immune combinations must be developed to treat unresponsive tumors. This task aims to validate at the single-cell level the role of ALK (Anaplastic Lymphoma Kinase) in the DNA damage response system, in order to select patients who could benefit from combinatorial therapy for preventing disease progression. In particular, ALK overexpressing cells will be characterized in 3D in vitro functional preclinical models, including both spheroids and organoids. The DNA damage, along with phenotypical features, will be analyzed under ALK overexpression and targeting by TKI. |
