Descrizione sintetica in italiano | The main goal of this project is the identification and characterization of genes potentially related to the phytoalexin-mediated defense mechanism in grapevine in response to stresses. The project will be articulated in four basic WP: WP1: Identification, annotation and phylogenetic analysis of all grapevine PDR genes predicted based on the 12X V1 assembly coverage of the PN40024 genotype. WP2: Screening of the expression of each member of the PDR sub-family in data-sets obtained from the mRNA-seq analysis to check the existence of other genes showing interesting induction patterns. Co-expression of candidate genes and selected VvSTS will be evaluated by quantitative RT-PCR in detailed time-courses performed on grapevine leaf discs treated under biotic and abiotic stresses. WP3: Study of the protein localization of candidate ABC transporters using GFP fusion constructs in heterologous and homologous systems. |

WP1: Bioinformatics analysis of the grapevine PDR subfamily based on predictions obtained from the 12X coverage assembly of the PN40024 genotype. Deduced PDR aminoacid sequences will be aligned using MAFFT software package and a phylogenetic tree will be construct using the Neighbor-Joining tree method. WP2: Selection of the most responsive PDR members and confirmation of co-expression with selected VvSTS genes by quantitative RT-PCR. Candidate ABC transporters will be selected and their co-expression with selected VvSTS will be validated by quantitative RT-PCR. WP3: Study of the protein localization of candidate ABC transporters using GFP fusion constructs in a heterologous (onion tissues) and homologous (grape protoplast) system. WP4: In planta functional validation for a role of candidate ABC transporters in resveratrol traslocation. The applicant will have to follow all the laboratory experiments and also to be responsible of the organization of all the programmed activities |
